MILLWALL boss Neil Harris will change most if not all of his first-choice starting XI at Kenilworth Road.
Steve Morison and Ben Thompson went off injured in the 1-0 FA Cup win over Southend last Friday but both would likely be rested against Luton.
Shane Ferguson is away on international duty with Northern Ireland.
Harris will again give the likes of Noah Chesmain (above), Sid Nelson and Tom King their chances to impress.
Shaun Cummings is also in line to make his first appearance since February, while Harry Smith and Paul Rooney could be rewarded for their good U23 form with first-team debuts.
Like Harris, Luton boss Nathan Jones could name a completely different line-up to the one that defeated Exeter 3-1 on Saturday, a result that leaves Luton in fourth place in League Two.
Jones is looking forward to a "stern test" as both sides, already qualified, contest top spot and a home draw in the last 32 of the competition.
"It's a great test" he told local media. "Millwall are a real traditional club and it will be a very difficult game.
"They've started to find their feet a bit now. They had sort of an average start, if you like, after last year's run (to the play-off final), they'll want to be there and thereabouts and I'm sure they will.
"Under Chopper, they'll do very well and they're starting to find their feet, so it will be a real stern test.
"We like testing ourselves. We enjoyed the test against Gillingham, we enjoyed the test we had against Aston Villa, Leeds (in the League Cup) and a different one from West Brom and every week in League Two. We're going to look forward to it."
